Estetrol (E4) w tabletkach Drovelis pełni rolę estrogenu. Jest to stosunkowo nowy - jeśli chodzi o wykorzystanie w medycynie - związek. W warunkach naturalnych wytwarzany jest wyłącznie przez wątrobę płodu podczas ciąży. Dlatego określany jest przymiotnikiem "native" - ludzki i naturalny. Dotychczas przeprowadzone badania kliniczne estetrolu w połączeniu z drospirenonem wykazały, że E4 ma minimalny wpływ na hemostazę, czynniki krzepnięcia, inhibitory krzepnięcia, fibrynolizę, angiotensynogen, triglicerydy i cholesterol. Estetrol może więc prezentować lepszym profil bezpieczeństwa niż inne, dotychczas stosowane estrogeny.
