Biologiczny okres półtrwania azytromycyny (składnika aktywnego leku Sumamed) jest długi i w końcowej fazie eliminacji może wynosić nawet 68 godzin, dlatego możliwe jest jej przyjmowanie tylko raz na dobę przez relatywnie krótki czas, na ogół 3-5 dni.

Stwierdzono, że stężenie terapeutyczne (czyli dosyć wysokie) azytromycyny w tkankach utrzymuje się jeszcze około 5‑7 dni po podaniu doustnie ostatniej dawki, natomiast całkowita eliminacja leku może zająć do około 17 dni.
