I know that the recommended dose is one ampoule of pen syringe, while my husband is to take a small dose, i.e. not 250 micrograms, but 25 - we don't want to spin this gentleman, so as not to waste the drug. Does anyone know how many doses of 25 micrograms are in one pen? Will it be one twist?

The Ovitrelle pen contains one full dose of 250 micrograms (0.5 ml), equivalent to ten doses of 25 micrograms.
The manufacturer does not provide information on the dose equal to one turn of the pen, because it is intended for single use and is not used to administer smaller doses.
Your doctor may recommend a lower dose off-label, according to your experience, in which case he or she should tell you how many times you need to turn the pen to get the desired dose and how to administer the injection.
