Severe cases of drug-induced liver injury, including liver cell damage, liver stasis, and acute hepatitis, have been reported with long-term use of Plaquenil.
Ototoxicity, i.e. damage to nerve elements of the inner ear, associated with the use of hydrodroxychloroquine is very rare but may be irreversible.
It is important to keep the daily dose as low as possible during long-term treatment.
The upper limit is 400 mg/day/year, corresponding to 6 mg/kg.
Patients on long-term treatment should have periodic blood tests (complete blood count) and if abnormalities occur, hydroxychloroquine treatment should be discontinued after consultation with a physician.
Please remember that Plaquenil is a prescription drug and any changes in its dosage or use in general must be made by the attending physician who knows your medical history and has the test results.
